Your challenge is to develop an operational sleep shift scheduling tool that provides autonomous customization of a schedule for sleep, exercise, and nutrition to manage fatigue.

How We Addressed This Challenge

This Tool uses a predicting algorithm working with great efficiency that uses data from a person's Microbiome , Gene Expression , Nutrition profile [what she ate? when did she eat this] , Exercise [ Parasympathetic nervous system Excitation and calories expelled], and Level of comfort [ 3D printed sound absorbers for the ISS] to predict REM sleep, NREM sleep, sleep duration etc.

Not just prediction it even tell's you how you can change your microbiome and if your sleep aid is effective, is lemon water at night for you more effective than benadryl? Should you exercise at Dusk rather than at noon? Should you eat a more meditteranian diet and increase protein intake? This way you body's sleep will be more easier to maintain with so many variables that you can change, just pick one that suits you!





GUT APPROACH

Microgravity changes in the Gut [1] have been linked in many experiments carried out by NASA and their partners with many Research Laboratories in the world. Such changes have been observed irrespective of duration in space in different statistical settings. There has also been a link of Microbiota changes with problems in sleep, circadian disturbance and even depression[2] These had not been linked before, and We believe can shed some interesting highlights.

I have designed an assay plate for long duration space travel, deep space travel, Moon base and even for possible missions to mars. It is cheap and effective can be given to general population as well.

It uses CRISPR and faecal matter collected from astronauts to check for microbiota changes. Quantification using measuring associated chromogen signal strength in Pixelated data feuded computer program to identify and quantify bacteria changes.

Cas13 and Cas12 will be used to identify specific bacteria in the faecal matter, if there is a match with the guide DNA/RNA for the respective enzymes, chromogen associated proteins will light up after TRAnS cleavage is activated and there will be detectable signal string in the plate. Dilations will be in place in the assay plate, and i10 high quality images of this plate will be fed to the computer program to identify quantity of the bacteria. This way it will be a fast and efficient analysis of gut bacteria and health.

Bacteria whose DNA have been predesigned to be tested:- Clostridiales, Lactobacillales, and Bacteroidales and Firmicutes

These have been investigated and cause chanages in the circadian clock genes gene expression and microbicidal microbiome structural changes in these structures. Quantifying the shift in shift workers and also people with jet lag.

The analysis will help the user to take specifically designed pro-biotics to bring their gut back to health, and also to help them understand foods to take to naturally bring back efficiency.

SCIENC Behind the technique:- CRISPR CAS12/13 bind to the template which they are made to recognise. If the sample has the certain DNA / RNA the enzyme starts to cleave any DNA / RNA it can find in its affinity- called trans cleavage. Certain reporter strands of DNA are present which one cleaved send out a color signal.

This signal is fed into a computer program that recognized the signal strength using pixel information and Dilution scores given estimates. These programs have been written for immunohistochemistry in C+, here we will use python for chromogenic Quantitative detection.

These will then asses micrbiota changes in microgravity causing circadian misalignment, anxiety and depression in astronauts.

This cheap assay can be used by anyone in the world, since the computer program will be freely available for analysis.



Nutrition TAP-BAND

A band that uses your movement, Hear beat and food intake to analyse when you should sleep.

Parasympathetic innervention and Carbohydrates, Bioamine, Fibre , Sugar , and Sodium analysis for sleep time and predicting NREM and REM sleep, sleep consistency. And methods for nutrition associated better sleep.



Hence NUTRIME-TAPBAND

on your tap the wrist band wakes up, you can click a picture using the small inbuilt camera and then using Machine Learning it Predicts the Sleep Predictor Value of the Mixed Food . The prediction is very effecient.

There is a substantial body of evidence to indicate a role of carbohydrate intake on sleep indexes (. Both high-carbohydrate (HC) and low-carbohydrate (LC) diets are associated with changes in sleep architecture

Meal Time Glycemic index.

The glycemic index (GI) of carbohydrates has also been studied as a dietary factor related to sleep architecture. The investigation of the effects of both GI and meal timing should be 4h before sleep time.


Mostly all food nutrition apps that count your calorific intake require you to put in the value yourself or have a small data set which can add values using the QR code. No app on the market is available for telling you the calorific value of home cooked food, a normal source of intake for many countries and 80% of rural population of the world.




Gene Expression:-


"Many astronauts reported taking Hypnotic drugs, sometimes even popping another one, although even one was recomended" This line made me think are the drug metabolism in space different?


DRUG METABOLISM AND GENE EXPRESSION OF MASTER REGULATOR GENES SLEEP REGULATORS


I propose looking at how each astronaut individual expression of genes BMLA1 and CLOCK and the polymorphisms in the sleep clock genes.

Especially important for deep space mission astronauts.


Drugs like Benadryl Zalpiedem and Zalpeopl and all naturally occuring foods causing increase in Biogeneric Amines will be also important in the analysis

Sound absorbers can be printed and their openScad code is available to view; These panels are Perforated panels with coiled air chamber , which allows low frequency humming noises of machines and causes sound absorber


these use printing micro holes in a cuboid structure and applying two 3D printed 1cm Mesh with each other overlapping and then apply them on a frame to mask the astronauts sleep pod.

17 feature points on the facial muscles are important and their activity predicts drowsiness Active appearance model is used to identify the 3D points in space it classified the expression into 4 levels.

ANSWER

Using 5 parameter a tool will help schedule time for perfect sleep and rate each person for their Insomnia and Circadian misalignment.


  1. Microbiome:- The CiMSP assay photographs when feeded into the computer program will rate the person from 1-5 for likely circadian missalignment due to microbiome chages. These cause the gut-brain axsis change.
  2. Nutrition :- Sugar , calories and protein mediation. Low protein - less sleep and High protein - maintain sleep a problem. So is your nutrition aligned for perfect sleep? The algorithm will rate it 1-5, for nutrition effected sleep.
  3. Exercise:- Parasympathetic Excitation rated 1-5. Inactivity in last 60 minutes, log checks etc can be done before astronaut down time.
  4. Gene Expression; Drug metabolism and other metabolites that effect sleep are studied and personal scores given for a genetic deposition to sleep dysregulation and with how are they overexpressed or underexpressed due to ongoing factors.
  5. Sound Absorbers:- cheap and effective and more comfortable that ear plugs. These can be attached like a cloth to the walls of the astronaut personal cabin.




We are a pair of high school and college student who want to make this challenge solution a reality. by the help of this incubator we want to launch Nutrime-Tap Band to the world for free and also make the CiMSP assay a reality. [care was given to design this assay and it works in microgravity - Patent Applied]


How We Developed This Project

Principles behind Idea?

neurotoxic metabolites such as D-lactic acid and ammonia produced by the gut microbiota may pass through the vagus nerve into the CNS, thereby affecting brain function, stress responses, and sleep structure there is shift experience changes circadian clock gene expression and microbial community structure probiotics rich in specific bacteria, such as Lactobacillus, Bifidobacterium, and Enterococcus, may cause changes in emotional and cognitive indicators by acting on the ENS and the immune system, thereby improving stress response and exerting an anti-anxiety effect

How We Used Space Agency Data in This Project

The lancet study Prevalence of sleep deficiency and use of hypnotic drugs in astronauts before, during, and after spaceflight: an observational study was how I used NASA data. I also used Data from the RT-PCR studies on the ISS and the 3D printer.
